Fixing Conditions in the Freeze Substitution Technique for Light Microscopy Observation of Frozen Beef Tissue
نویسندگان
چکیده
The freeze substitution histological technique allows the indirect light microscopicobservation and the quantitative evaluation of ice crystal size in frozen tissues. Theuseofchemical fixatives in substitu ting fluids improves morphological and histochemical preservation of the tissue. Fixation conditions become important since this step can introduce modifications in crystal sizes . Effects of temperature on: a) diffusion rate of fixing solution in tissue, b) recrystallization rate of ice in frozen beef and c) variations of the froze~ water fraction were ana l yzed, establishing that 1sothermal freeze fixatio n constitutes an appropriate method for histological observation. Fixing at temperatures lower than that of the sample, in an attempt to reduce recrystallization effects , involves an increase of the frozen water f~action i.n the tissue leading to modifications in s12e of 1ce crysta ls and to changes in the histological structure. Spray-freezing freeze substitution (SFFS) of cell suspensions for improved preservation of ultrastructure.
Some unicellular organisms present challenges to chemical fixations that lead to common, yet obvious, artifacts. These can be avoided in entirety by adapting spray-freezing technology to ultrarapidly freeze specimens for freeze substitution. To freeze specimens, concentrated suspensions of cells ranging in diameter from 0.5-30 pm were sprayed with an airbrush at 140-200 kPa (1.05-1.5 torr; 20.3...
متن کاملMethods and Principles of Fixation by Freeze-Substitution
Freeze-substitution is based on rapid freezing of tissues followed by solution ("substitution") of ice at temperatures well below O degrees C. A 1 to 3 mm. specimen was thrown into 3:1 propane-isopentane cooled by liquid nitrogen to -175 degrees C. (with precautions). The frozen tissue was placed in substituting fluid at -70 degrees C. for 1 week to dissolve ice slowly without distorting tissue...
متن کاملAn alternative means of retaining ocular structure and improving immunoreactivity for light microscopy studies
PURPOSE Several properties of ocular tissue make fixation for light microscopy problematic. Because the eye is spherical, immersion fixation necessarily results in a temporal gradient of fixation, with surfaces fixing more rapidly and thoroughly than interior structures. The problem is compounded by the fact that the layers of the eye wall are compositionally quite different, resulting in diffe...
متن کاملA Study of Extracellular Space in Central Nervous Tissue by Freeze-substitution
It was attempted to preserve the water distribution in central nervous tissue by rapid freezing followed by substitution fixation at low temperature. The vermis of the cerebellum of white mice was frozen by bringing it into contact with a polished silver mirror maintained at a temperature of about -207 degrees C. The tissue was subjected to substitution fixation in acetone containing 2 per cent...

In this study, to mimic the mineral and organic components of natural bone, hydroxyapatite[HA] and gelatin[GEL] composite scaffolds were prepared using the solvent-casting method combined with a freeze drying process. Glutaraldehyde[GA] was used as a cross linking agent and sodium bisulfite was used as an excess GA discharger. Using this technique, it is possible to produce scaffolds with mecha...
